He began his basketball journey at Lockport Township High School, where he quickly stood out. In college, he first played at Moraine Valley Community College (2011–12) before transferring to Bowling Green State University, from which he graduated in the summer of 2015.
During his three seasons at Bowling Green, Holmes made history as the only player in the program’s history to record more than 1,000 points, 600 rebounds, and 200 blocks. He also became the school’s all-time leader in blocked shots, with a total of 244.

His outstanding final season in the NCAA, in which he averaged 14.7 points, 8 rebounds, and 2.7 blocks per game, earned him MAC Defensive Player of the Year honors and a place on the All-MAC First Team. Holmes spent three seasons with the 76ers, averaging 7.4 points, 4.2 rebounds, and 1 assist per game, while also appearing for the Delaware 87ers, the team’s G-League affiliate.

He subsequently played four seasons with the Sacramento Kings, posting averages of 10.4 points, 6.6 rebounds, 1 assist, and 1.1 blocks per game. The 2020–21 season was the best of his career, as he finished the regular season with 14.2 points, 8.3 rebounds, and 1.6 blocks per game.

During his time with Sacramento, Holmes delivered the most notable performances of his career. On December 4, 2019, he scored 28 points against the Portland Trail Blazers, while on November 5, 2021, he recorded a double-double with 23 points and 20 rebounds against the Charlotte Hornets.

Additionally, in three separate games wearing the Kings jersey—January 11, 2021 (vs. Indiana Pacers), January 22, 2021 (vs. New York Knicks), and February 1, 2021 (vs. New Orleans Pelicans)—he registered 6 blocks in a single game.

In the 2023–24 season, Holmes began the year with the Dallas Mavericks, averaging 8.2 points, 4.7 rebounds, and 1.1 blocks per game. He later joined the Washington Wizards, which marked the final stop of his NBA career. With Washington, he averaged 7.3 points, 5.9 rebounds, and 1.1 assists per game.
